DESCRIPTION
Selegiline is a selective, irreversible inhibitor of monoamine oxidase B (MAO-B), used chiefly for Parkinson’s disease and, at higher doses, for major depressive disorder. It works by enhancing levels of dopamine and, at increased doses, serotonin and norepinephrine in the central nervous system.
Selegiline binds to and inactivates the MAO-B enzyme, which is present in high concentrations in regions of the brain associated with movement and mood. By inhibiting MAO-B, it blocks the breakdown (oxidative deamination) of dopamine, allowing higher levels of dopamine at synaptic sites in the basal ganglia and other brain regions. When given at higher doses, selegiline can also inhibit MAO-A, raising levels of serotonin and norepinephrine.
Selegiline improves movement and mood by increasing dopamine availability in Parkinson’s and, at higher doses, broadens monoaminergic neurotransmission to include serotonin and norepinephrine for depression therapy.
Benefits
- Improves movement and muscle control in Parkinson’s disease.
- May delay disease progression and extend levodopa efficacy.
- Provides modest antidepressant effects, comparable to other antidepressants.
