DESCRIPTION
Amitriptyline is an antidepressant tricyclic class of compounds derived from dibenzocycloheptadiene. The mechanism of antidepressant action is associated with an increased concentration of noradrenaline (noreprinephrine) in the synapses and/or in the central nervous system due to reverse neuronal capture of these mediators. After long-term use, Amitriptyline produces a reduction in brain beta-adrenergic and serotonin receptors function, normalizes adrenergic and serotonergic transmission, restores the balance of these systems, impaired in depressive states. This medication helps to reduce anxiety, agitation and depressive symptoms in patients with anxiety-depressive disorders. Amitriptyline has also an analgetic effect associated, as it believed, with changes in the concentration of monoamines in the central nervous system, especially serotonin, and the influence on the endogenous opioid system. INSTRUCTIONS & DOSAGE
Active substance: amitriptyline hydrochloride 25mg

Amitriptyline therapy is usually started at 25-50mg at bedtime and the dose is individually increased during the next 5-6 days up to a maximum of 150-200mg/day divided in to 2 parts (the highest part at bedtime). If there are no noticeable improvements during the second week of therapy the dose is recommended to be increased up to 300mg/day. After a therapeutic effect in treating depression has been achieved, the dose is reduced to 50-100mg/day and maintained for not less than 3 months.

Elderly people will require the dose of 25-100mg/day to be taken at bedtime. After a therapeutic effect has been achieved, the dose is reduced to 25-50mg/day.

Bedwetting, age 6-10 years: 10-20mg/day at bedtime.

Bedwetting, age 11-16 years: 25-50mg/day

STORAGE
Store in dry place at room temperature. Do not exceed storage temperature higher than 25 degrees Celcius. Keep away from direct sunlight. Keep locked and away from children.
